Base yourself near the River Walk or downtown so you can walk to most sights; use rideshare (Uber/Lyft) for Natural Bridge Caverns, the missions, and any late-night returns rather than renting a car for the whole weekend. For the missions, you can also use the VIA VIVA bus routes if you prefer public transit.
